Dynamics of the mitochondrial outer membrane proteome in mitophagy and apoptosis
Accurate detection and quantitative measurement of mitochondrial proteins and their modifications is crucial for understanding mitochondrial function. Current shotgun proteomics workflows are able to cover and quantify the MOM proteome to a high extent and enable comprehensive detection and quantification of posttranslational modifications, which is critical for fully understanding the function of MOM-associated proteins, such as the PINK1 kinase or the PARKIN ubiquitin ligase. In this project, we will use stable isotope labeling to perform relative and absolute quantification of the MOM-associated proteome, phosphoproteome and ubiquitylome in mammalian cell lines undergoing apoptosis and/or mitophagy.
