VDAC as an intersection of the mitophagy pathway
The major recessive Parkinson’s disease genes encoding the kinase PINK1 and the ubiquitin ligase parkin mediate the autophagic elimination of damaged mitochondria. We will employ mass spectrometry to systematically identify post-translational modifications of MOM proteins throughout the time course of mitophagy. As VDAC is known to be an important attachment MOM protein of autophagy-targeting ubiquitinylations we will more specifically investigate this protein with emphasis on the VDAC interactome. VDAC is not only involved in mitophagy, but also in apoptosis. We address VDAC at the crossroads of normal mitochondrial function, mitochondrial quality control and cell stress.
